HOW TESTOSTERONE WORKS Once testosterone has been made by the endocrine system, it is secreted into the bloodstream and circulated throughout the body, finally arriving at its target cells. To exert its effects, testosterone must enter its target cells. The outer cellular membranes (walls) of cells are made of lipids and proteins - called the lipid bilayer. This layer is selectively permeable. As a sex steroid with a basic structure of four fused carbon atom rings, testosterone is highly lipophillic and highly soluble (dissolvable) in lipids. It can pass in and out of lipids very easily. For this reason, once at its target cells, testosterone diffuses across the cellular membrane. This is sometimes called passive transport or non-mediated transport - because unlike protein hormones or amine hormones that need energy to enter the cell, testosterone requires no energy to diffuse across the membrane, instead diffusing because of chemical gradients. After diffusing the membrane, testosterone enters cellular cytoplasm and binds to testosterone specific steroid receptors (called androgen receptors) inside of the cell. These receptors and some proteins (called steroid receptor co-factors) are needed for the cell to activate and for the next step to occur. Once it has bound with the steroid receptors in the target cells, and provided that the steroid receptor co-factors are present, testosterone goes one step further and binds to the nucleus of the cell. Binding to the nucleus of the target cell involves testosterone binding to specific sequences of chromosomal DNA (hormone response elements). At this point, two stages of change occur transcription and translation. When testosterone binds to the cells nucleus, the specific sequences of chromosomal DNA must be decoded. They are copied onto RNA and direct protein synthesis. This process is called translation - literally translating the instructions from testosterone and producing a result. WHAT TESTOSTERONE DOES Testosterone is powerful and causes massive changes all throughout the body. Testosterone increases endurance by allowing you to work harder, longer. Testosterone increases recovery speed from exercise by increasing glycogen uptake, protein synthesis and the speed of muscular repair. Testosterone burns fat by liberating fatty acids from adipose tissue and increasing the activity of your fat burning machines, the mitochondria. HORMONAL ENEMIES OF TESTOSTERONE As strong as testosterone is, and as important and impressive as your endocrine system is at producing it, the endocrine system also produces many other hormones that limit testosterone levels and limit your gains in the gym. The hormones that work against testosterone are Estrogens SHBG Insulin Glucagon Cortisol Estrogens are made a number of different ways, but the most common is the enzymatic conversion of testosterone into estradiol and the conversion of androstenedione into estrone. Estrogen is the dominant female hormone, and while some estrogen in males is needed for normal health, high estrogen levels can cripple testosterone levels, stop muscle growth, cause water retention, weakness and cause unwanted fat gain. SHBG (Sex Hormone Binding Globulin) is made and secreted by the liver and while it doesnt affect your overall testosterone levels, it does limit the amount of metabolically active - useful - testosterone that you have. It does this by binding to the testosterone and rendering it virtually useless. For this reason, SHBG is also called androgen-binding protein. Insulin is made and secreted by the pancreas and, as previously mentioned, it is secreted by the pancreas when you eat carbohydrates. Insulin has a number of beneficial effects, but it becomes an enemy of testosterone when it spikes and is chronically high. In this situation, testosterone levels plummet, body fat gain occurs and testosterone levels stay depressed. Glucagon is also made and secreted by the pancreas and, like insulin, can be an enemy of testosterone depending on the environment within the body. Under certain conditions, glucagon is catabolic -muscle wasting - and causes the breakdown of muscle tissue for amino acids to fuel glucose manufacture. This is detrimental to testosterone levels and increases cortisol in response to inflammation. Cortisol is a stress hormone thats made from progesterone and is secreted by the adrenal glands. Cortisol is classified as a catabolic - muscle wasting - hormone. It is secreted in large amounts immediately following exercise. While low cortisol levels are acceptable, high cortisol levels will send your testosterone levels plunging. As a result, youll lose muscle mass, strength, your exercise recovery will slow down and youll be more susceptible to injuries. Unfortunately, a high amount of testosterone is bound by SHBG and left metabolically inactive. Therefore, while total testosterone levels are important, free testosterone levels are a measure of how much testosterone your body can use to build muscle and burn fat, and thus free testosterone levels determine your results in the gym. To build muscle you must Increase your total testosterone levels Maximize your free testosterone levels Unfortunately, Figure 2 shows that while testosterone production levels peak at age 18, they begin a steady decline at around age 20 and continue to decline over the course of your life. By age 65, your testosterone levels decline to less than half what they used to be. Compounding this drop in total testosterone production, free testosterone levels decline even more dramatically as your total testosterone production drops off. And, to make matters worse, your body increases its production of estrogens, cortisol and other muscle destroying hormones this all results in less muscle, weakness and more body fat.
